Het is het jaar 2046. Na een vernietigende oorlog wonen mensen en aliens samen op aarde. Veteraan Joshua Nolan keert terug naar de ruïnes van zijn geboorteplaats St. Louis, waar inmiddels het stadje Defiance verrezen is. Daar probeert Nolan als sheriff de orde te handhaven.

Following the events of the season two cliffhanger finale, season three of Defiance opens on the eve of an impending military siege upon the town, which has fallen on hard times following the collapse of its lucrative mines.

    Nolan and Irisa are mysteriously rescued from their underground confinement just in time to discover that enemy forces are marching on the town of Defiance, which has fallen on hard times following the collapse of the gulanite mines. Meanwhile, Datak, Stahma and Rafe run into deadly complications on their quest to reunite their family; and Alak comes to realize just how unstable her mother-in-law is during their journey through the woods.
