Een samoerai heer heeft de organen van zijn pasgeboren zoon geruild voor achtenveertig demonen in ruil voor dominantie op het slagveld. Toch overleeft het verlaten kind dankzij een medicijnman die hem uitrust met primitieve protheses - dodelijke protheses waarmee de verongelijkte zoon de veelheid aan demonen zal opjagen om zijn lichaam stukje bij beetje terug te winnen, voordat hij zijn vader confronteert. In Japan's Warring States period, Lord Daigo Kagemitsu makes a pact with 12 demons, offering them his unborn son in exchange for the prosperity of his lands. Years later, young thief Dororo is attacked by a ghoul but rescued by a mysterious boy, whose body parts turn out to be almost entirely prosthetic.
